The mouse evidence: a rare compression of morbidity

The pivotal preclinical study came from the Buck Institute for Research on Aging in 2020, published in Cell Metabolism. Researchers fed calcium-AKG (CaAKG) to aging mice and tracked them across their remaining lives. The animals did not merely live a little longer; they lived better. AKG reduced frailty, lowered a panel of inflammatory markers, and produced what gerontologists prize most — a "compression of morbidity," meaning the sick, frail portion at the end of life grew shorter rather than simply being stretched out.[2]

The eight-year headline — and why to read it carefully

Then came the number that launched a thousand supplement orders. In 2021, a retrospective study published in the journal Aging examined 42 people who had been taking Rejuvant, a delayed-release calcium-AKG formulation combined with vitamins A and D. Using a commercial DNA-methylation age test, the researchers reported that, after an average of about seven months, participants showed a mean reduction in biological age of nearly eight years.[4] For a single, well-tolerated supplement, an eight-year rollback of the epigenetic clock is a spectacular figure — and it spread accordingly.

Here the discerning reader must slow down, because the design of that study carries weight the headline does not. It was retrospective and uncontrolled: there was no placebo group, the participants had chosen to buy and take the product themselves, and people who feel a supplement is working are more likely to keep taking it and to volunteer their data. Self-selection of this kind can manufacture impressive-looking numbers on its own. A subsequent, larger analysis of AKG users reported a more sober reduction of roughly 1.8 years in biological age — still a positive signal, but a very different order of magnitude, and a useful lesson in how the first, boldest number in a young field is rarely the one that survives.

The honesty clause: what is still missing

An article that stopped at the eight-year figure would be selling you a headline rather than the truth. The candid state of the evidence in 2026 is this: the animal data for AKG is genuinely strong and mechanistically coherent, but the human data that everyone quotes is not yet the kind that settles questions. The elegant review of AKG's biology in humans, published in Trends in Endocrinology & Metabolism, reached exactly this measured verdict — the pathways are compelling and the older surgical-nutrition safety record is reassuring, but rigorous longevity trials in people had not yet been done.[5]

The good news is that the missing piece is being built. Randomised, double-blind, placebo-controlled trials of calcium-AKG against biological-age endpoints are now underway, and they are the only instrument that can convert an intriguing retrospective signal into a trustworthy fact. Until they report, the correct posture is neither dismissal nor enthusiasm but patience: AKG has cleared bars most supplements never approach, yet its most exciting human claim still rests on evidence too soft to bear the weight placed on it.
